Abstract

Background. More than 500000 people worldwide have bacterial keratitis every year. Conservative therapy with antibacterial drugs often leads to the emergence of resistant strains of bacteria, allergic reactions and complications. Silver nanoparticles (SNP) are current for the role of alternative to modern antibiotics. Objective. To investigate the effect of applications of therapeutic films with SNP sizes of 30 nm, obtained by citrate (CM)and electron-vacuum-beam methods (EVBM), on the ultrastructure of the epithelium and stroma of the rabbit cornea after modeling in animals of moderate bacterial keratitis. Methods. Bacterial keratitis was modeled for all experimental animals, then equally divided into 4 groups: 1 control group, 2 group received applications of films with SNP, received CM on the cornea, group 3 – films with SNP obtained by EVBM, group 4-films with decametoxin. Corneal fragments were taken for electron microscopic examination. Results. Films with silver nanoparticles with a size of 30 nm obtained by CM have the most effective therapeutic effect on the regeneration of rabbit cornea tissue after simulated bacterial keratitis of moderate severity. Help to reduce the exudative phase of the inflammatory process, enhance the proliferative response, a delicate collagen skeleton is actively formed. After application of films containing silver nanoparticles 30 nm in size, obtained by EVВM, signs of inflammation in the tissues remain, formation of cells of the anterior epithelium and connective tissue skeleton of the cornea stroma proceeds slower than after the exposure of films containing SNP, the resulting CM. After exposure to films with decametoxin, the restoration of the anterior corneal epithelium and stroma is slowed down, signs of the exudative phase of the inflammatory process are expressed. Сoarse-fibrous connective tissue is formed in the stroma. Conclusions. The effectiveness of experimental therapy of bacterial keratitis in rabbits increases in the series: films with decametoxin → films with SNP obtained by EVBM → films with SNS obtained by CM.
